چکیده

Introduction Natural Killer Cell activity has traditionally been assessed by detecting the lysis of tumor cells in The Chromium Release assay. This classic assay relies on radioactive labels, is laborious, and requires substantial quantities of patient blood. Moreover the Chromium Release assay provides low signal to noise ratios. We have developed an assay that can visualize individual target cells to detect cytolytic activity within a high signal to noise range, without involving radioactivity, via high-throughput imaging. We also developed a miniaturized version of this assay to measure NK activity with one tenth of the blood needed for a classic Chromium Release assay, and with less labor.

Hyperthermia Increases Natural Killer Cell Cytotoxicity against SW-872 Liposarcoma Cell Line

Background: Although there is convincing data in support of the effectiveness of hyperthermia in tumor therapy, the molecular mechanisms underlying the clinical effects of hyperthermia are still poorly understood. Objective: To investigate natural killer (NK) cell cytotoxicity against heat-treated SW-872 and HeLa tumor cell lines. Methods: NKG2D ligands and HLA class I transcription were examin...

THE CYTOTOXICITY PATHWAY OF NATURAL KILLER CELLS IN CORD BLOOD COMPARED TO P ERIPHERAL BLOOD

The cytotoxic activity of natural killer cells is usually tested by radioactive assay CSI Cr release assay), which detects the release of cytoplasmic contents after plasma membrane disintegration of dying cells. In contrast to this indirect evaluation of cytotoxicity, the assessment of cell damage by flow cytometry aims to provide a more exact characterization of the death pathway via detec...

Measuring Cytotoxicity by Bioluminescence Imaging Outperforms the Standard Chromium-51 Release Assay

The chromium-release assay developed in 1968 is still the most commonly used method to measure cytotoxicity by T cells and by natural killer cells. Target cells are loaded in vitro with radioactive chromium and lysis is determined by measuring chromium in the supernatant released by dying cells. Since then, alternative methods have been developed using different markers of target cell viability...

Increased Natural Killer Cells Activity in Schizophrenic Patients

Background: Schizophrenia has been associated with altered immunity. Different studies regarding natural killer cell activity (NKA) in schizophrenic patients have shown inconsistent results. Objectives: To evaluate NK cell activity in schizophrenic patients in comparison with healthy control individuals.   Methods: 30 medication-free schizophrenic patients and 41 healthy sex, age and smoking st...

Use of flameless atomic absorption spectroscopy in immune cytolysis for nonradioactive determination of killer cell activity.

We describe here a novel method to evaluate natural killer (NK) cytolytic activity by use of flameless atomic absorption spectroscopy (GF-AAS). This technique may be adopted for use in laboratories equipped with electrothermal atomic absorption spectrometers. Nonradioactive Cr as Na2CrO4 was used to label target cells (K562), and cell lysis was evaluated by measuring Cr released after 4 h of in...
